高劑量稀釋裝置應用於豬瘟疫苗的施打

在此研究中,我們針對高劑量單位豬瘟疫苗的稀釋技術進行開發。利用先前所開發並已獲得之高劑量稀釋裝置專利為基礎,針對此高劑量的豬瘟疫苗在稀釋與使用施打上的需求進行裝置配備的改良與評估,以尋求最佳使用裝置。在所設計的五種裝置配備中,以第五型裝置使用性最佳,只需一次動作即可混合完畢,且沒有因瓶內壓力不足而有混和不良的問題。針對第五型裝置所稀釋的疫苗液進行疫苗穩定性測試。其結果顯示,高劑量疫苗分別在37℃與4℃下於裝置內進行稀釋然後靜置3小時後,每一劑量的病毒力價仍可維持104TCID50/ml力價。此第五型稀釋裝置能成功地運用於高劑量單位豬瘟疫苗的稀釋,不僅使用方便,且能維持疫苗力價的穩定至少3小時,使凍結乾燥疫苗得以在現場快速無菌回溶以供馬上使用施打,提供農民在大量施打豬隻時的便利。

英文摘要

In this study, devices for high-dose units of reconstitution of classic swine fever virus (CSFV) vaccine were designed. On the basis of the patentedhigh-dose dilution device, the devices with outfit were designed and reformed for optimal use. Among thereformed five devices, the type 5 device was the best one due to its ease to mixing with a single action and without the problem of poor mixing. The vaccine stability after reconstitution under the type 5 device was examined. The results showed that the high-dose units CSFV vaccine reconstituted under the device type 5 either at 37 C or 4 C for 3h incubation, the viral titer of 104 TCID50/ml in each dose could be kept. The type 5 device was successfully applied to the reconstitution of high- dose units freeze-drying CSFV vaccine in this study. This device was not only easy to use, but also could maintain the stability of the vaccine for at least 3 hours after reconstitution. That provides the convenience for farmers to reconstitute vaccine aseptically and quickly in the field for immediate use when a large number of pigs need to the vaccinated.
